Vertebrate muscles It is among the vertebrates that the most exhaustive studies of fibre diversity have been carried out. The successive levels of investigation, from the biochemical, through the electrophysiological to the mechanical, show just h o w fundamental are the differences between fibres. Amphibian skeletal muscle has been examined at all of these levels and The Fine Structure of Skeletal Muscle 43 forms an excellent introduction to muscle fibre heterogeneity. The early work of Tasaki and Mizutani (1944), Kuffler and Gerard (1947), Kuffler and Vaughan-Williams (1953), Burke and Ginsborg (1956) and Orkand (1963) showed the presence of two distinctive types of fibre in frog skeletal muscle, fast and slow.

Such an extensive system of internal tubules acts to greatly increase the muscle fibre surface area, and Falk and Fatt (1964) have suggested that much of the fibre membrane capacitance may be located in the walls of the T system. One would thus expect to see some correlation between the extent of T system development and the total muscle fibre membrane capacitance, but this topic has not so far attracted much attention (see Peachey, 1965; Selverston, 1967). The triad and dyad At discrete loci the T tubules make distinctive contacts with elements of the SR, and it is the arrangement of the opposed profiles which is responsible for the two different names used (Fig.

This gives the desmosomal area a 'beaded' appearance, for which the term 'septate desmosome' has been coined. The nexal regions of the disc are formed by fusion of the outer membrane leaflets with a total abolition of the extracellular space at that point. These parts of the intercalated disc are shown somewhat diagrammatically in Fig. 5. Barr et al. (1965) have examined the structure of the intercalated disc in cardiac muscle exposed to media of varying osmotic strength. In hyperosmotic media the cells became shrunken and the intercellular gaps between the adjacent cell membranes along the disc were widened, and, more importantly, separation of the nexal membranes occurred.
